Mutation of DYS385b=11 to 12

The diagram below shows that all the descendants of John Ferguson b.~1726, VA have DYS385b=12 whereas the other descendants of John Ferguson b.~1682, VA have DYS385b=11. This is one of the more important results in the project for it provides a definitive tool for confirming descendants of John Ferguson b.~1726, VA and Mary Gill.
Mutation

Mutation of CDYb=37 to 36

The haplotypes of all the persons who tested in the above diagram are shown below. Notice that everyone with DYS385a=11 has CDYb=37 and that everyone with DYS385a=12 has CDYb=36. This means there had to be a mutation of CDYb at the same time DYS385a mutated. The probability of two single step mutations occuring in a father-son event is low but it happens. In a study by Decker et al, they were able to observe twenty four differences between father and son and in that set the were two instances of two single step mutations. That gives a ball park figure of 8% strated as: when a father son difference exists, 8% of the time in involves two single step mutations. That ball park estimate will likely be higher in cases involving CDY since it mutates about ten times faster than any of the 17 markers used in that study. The bottom line is that in the present example the observed DNA results and genealogical relationships are consistent with one another. If a more severe test is required then all involved should upgrade to 111 markers. We have the expectation that inferred father son difference involving DYS385 and CDY would not differ on any of the additional markers, if that in fact is true then confidence in the genealogy would be very high.
